NSI-189 Freebase

NSI-189 Freebase – Product Information

NSI-189 is a unique investigational pharmaceutical chemical produced by Neuralstem, Inc. that has received interest in the nootropic community for its potential to improve mood, energy levels, and cognitive performance.

Animal studies suggest that NSI 189 could be a potential treatment for major depressive disorder (MDD) due to its alleged antidepressant effects. This experimental compound could possibly treat cognitive impairments or neurodegenerative disorders.

NSI-189 free base is a novel molecule that inhibits glucocorticoid receptors, distinguishing it from other antidepressant candidates (primarily serotonergic). It was tested because of findings of decreased hippocampal volume in depressed patients, which NSI-189 could theoretically treat due to its brain-derived neurotrophic factor. NSI-189 freebase products are also sold as pharmaceutical raw materials or pharmaceutical intermediates that are used for a specific multiple-dose escalation study.

Because NSI 189 freebase is considered an experimental chemical compound, it is, therefore, not safe for human consumption and should not be sold as a therapeutic agent. PureRawz sells NSI 189 for research use only.

What is NSI-189 Freebase?

NSI 189 is a small molecule that exhibits pleiotropic properties. It promotes neurogenesis in the hippocampus by attracting neural stem cells.

It is a neurogenic molecule being studied as a possible treatment for behavioral improvement during depression and neurodegenerative diseases.

The neurogenesis of the hippocampus region of the brain, which controls many important cognitive functions, is aided by NSI 189. Trials to investigate it as an alternative for those who have not responded to traditional medicines are still ongoing.

Possible Applications of NSI 189 Freebase

NSI-189 Freebase and Major Depressive Disorder

A neurogenic compound called NSI-189, benzylpiperizine-aminiopyridine, promotes neurogenesis in mouse and human neural stem cells derived from the hippocampus.

After daily oral administration for 28 days, it exerts behavioral efficacy at three different doses in a mouse model of depression. [R] [R]

The dentate gyrus neurogenesis and hippocampal volume in the treated mice’s brains both significantly increased. [R] [R]

In a small double-blind, placebo-controlled study, NSI-189 demonstrated significant promise as a treatment for MDD and simultaneously improved a cognition scale. [R] [R]

NSI-189 Freebase and Pro Cognitive Efficacy

Preliminary proof-of-concept studies show both antidepressant and procognitive effects, according to experts. NSI-189, when taken as a whole, is a novel multi-domain treatment capable of neurostructural benefits that may be used therapeutically to treat various neurodegenerative conditions. [R]

NSI-189 Freebase and Stroke

In adult Sprague-Dawley stroke rats, oral administration of NSI-189 beginning at 6 hours after middle cerebral artery occlusion and continuing every day for the following 12 weeks led to a significant improvement in stroke-induced motor and neurological deficits, which was maintained up to 24 weeks post-stroke. [R]

Stroke brains from NSI-189-treated animals underwent histopathological analysis. The results showed significant increases in neurite outgrowth as shown by MAP2 immunoreactivity, primarily found in the hippocampus and partially in the cortex. [R]

These findings imply that NSI-189 actively stimulated brain remodeling following stroke. Parallel in vitro investigations into this remodeling process revealed that oxygen-glucose deprivation and reperfusion (OGD/R) triggered common cell death processes that were then reversed by NSI-189 treatment, which was characterized by a consequential attenuation of OGD/R-mediated hippocampal cell death. How Does NSI-189 Freebase Work?

It is thought that NSI-189 has neurogenic properties that could aid in the treatment of depression and other neurological conditions. It functions by promoting neurogenesis, the process of creating new neurons in the brain.

This procedure helps to repair the connections between broken brain cells and develop new communication pathways. Histopathological evaluations, which involve a microscopic examination of tissue samples to ascertain whether cells or tissue structures have been altered as a result of cognitive impairment, trauma, infection, or disease, have demonstrated this.

When adult rats were given NSI-189 orally starting at 6 hours after middle cerebral artery occlusion and every day after that for the following 12 weeks, there was increased neurite outgrowth as shown by MAP2 immunoreactivity, according to a study.

In brain regions that a stroke or other injury has impacted, NSI-189 is thought to promote neuron growth and repair. This process is achieved by increasing the ability of existing neurons to produce new ones.

Expert opinion indicates that NSI-189 may not only affect neuron growth but also protect already damaged neurons from further harm. This seems to be accomplished by its antioxidant properties, which can lessen oxidative stress brought on by toxins or inflammatory reactions brought on by trauma or neurological disorders in the brain.

A particular clinical trial also shows that NSI 189 may pass the blood-brain barrier. [R]

Frequently Asked Questions

What are the potential benefits of NSI 189 freebase to cognition?

This novel compound is believed to be beneficial not only in treating depression but also in providing significant cognitive function. NSI-189 has been shown to stimulate neurogenesis in the hippocampus, improving learning and long-term memory.

What is the difference between NSI-189 freebase and its phosphate counterpart?

Pharmacologically, freebase NSI-189 can pass through membranes more efficiently, allowing it to be absorbed sublingually more quickly in research subjects. It is more potent in terms of weight.

NSI-189 phosphate, on the other hand, is salt, less potent by weight, and more readily absorbed orally. Several clinical studies are centered on phosphate salt due to its oral absorption qualities; however, the effects are essentially the same apart from absorption rates.

The harsh taste of NSI-189 phosphate can make sublingual administration uncomfortable for laboratory animals.
