Coluracetam

Coluracetam is a nootropic belonging to the racetam family. Racetams are a class of compounds that have the same chemical backbone (a pyrrolidone nucleus). Although their exact mechanism of action remains unknown, some racetams appear to modulate neurotransmitters. [R][R]

Coluracetam was originally developed in 2005 as a potential treatment for Alzheimer’s disease. Later on, it was in phase 2 clinical trials for the treatment of anxiety and depression. However, clinical research was discontinued after it failed to give conclusive results in both trials. [R]

However, preclinical studies have continued. These studies have shown that Coluracetam may reverse the symptoms associated with schizophrenia.

The nootropic has also been found to decrease anxiety in rats by 20%. In the same study, Valium reduced anxiety by 12%. [R][R]

How does Coluracetam Work?

Although its exact mechanism remains unknown, here are the reported effects:

Increases High-Affinity Choline Uptake

In a rat study, Coluracetam reportedly enhanced high-affinity choline uptake (HACU). This is significant as HACU is the rate-limiting step in the production of acetylcholine. This was also shown in the study in which, after enhancing HACU, Coluracetam facilitated acetylcholine synthesis. [R]

Binds to AMPA Receptors

Coluracetam has been proven to be an AMPA receptor agonist. AMPA receptors are critical for mediating glutamatergic synaptic transmission and plasticity, thus potentially playing a major role in molecular mechanisms underlying memory and learning. [R][R]

Shields NDMA Receptors from Glutamate Toxicity

In one preclinical study, Coluracetam was found to protect the NDMA receptors from glutamate toxicity. It has been shown that damage to these receptors is involved in traumatic brain injury, Alzheimer’s disease, stroke, and other brain diseases. [R]

Coluracetam Research

What follows is a summary of what clinical, animal, and cell-based research has to say about Coluracetam. The information given in this section is for informational purposes only and doesn’t constitute medical advice. PureRawz doesn’t sell Coluracetam for human consumption.

Coluracetam and Treatment Resistant Depression

A clinical study set out to investigate the effects of Coluracetam on 101 people with depression. All of the study’s participants also had anxiety and had failed treatment with 2 antidepressants. Coluracetam was found to have a potentially positive impact on depression at thrice daily 80 mg doses. [R]

However, the above study remains the only published piece of clinical research on Coluracetam. While the nootropic did undergo phase 2a clinical trials on major anxiety and depression, the results of those studies were never published. Clinical research on this compound has been discontinued.

Coluracetam and Memory & Learning

In a preclinical study, Coluracetam improved memory and learning in rats previously treated with a nerve toxin that hurts choline uptake in the brain. Notably, the improvements lasted beyond treatment and weren’t seen in rats that weren’t treated with the nerve toxin. [R]

In another rat study, Coluracetam treatment reversed the damage to the learning function caused by the recreational drug PCP. This compound inhibits choline acetyltransferase (ChAT), the enzyme that produces acetylcholine. Acetylcholine plays a role in memory and learning. [R][R]

Coluracetam and Cognitive Enhancement

A preclinical study saw repeated administration of Coluracetam to rats with chemically impaired memory. Results from the study show that Coluracetam brought about long-lasting cognitive enhancements by changing the choline transporter regulation system. [R]

Coluracetam and Anxiety

Results from a rat study indicate that Coluracetam treatment brought about a 20% improvement in anxiety. Significantly, the improvement in anxiety was 8% greater than the effect a single dose of Valium had on the animals’ anxiety levels in the same study. [R]

Coluracetam and Schizophrenia

In rats with nerve damage, Coluracetam was shown to increase the activity of ChAT, an enzyme that is impaired in schizophrenia. However, in the absence of large-scale, randomized clinical trials, one cannot be fully sure of the effects of the nootropic on schizophrenia. [R]

Coluracetam and Neuroprotection

Coluracetam may also help protect the brain from glutamate neurotoxicity induced by ionomycin, per results obtained by a preclinical study. Glutamate toxicity can cause various neurodegenerative disorders, such as Parkinson’s disease. [R]
