Behemoth Stack (S4 + LGD-4033 + MK-677)

What is Behemoth Stack (S4 LGD-4033 MK-677)?

Research has shown that aging leads to loss of muscle mass, strength, and leg power. This is mainly due to the loss of type II muscle fibers. Sarcopenia increases the risk of falls, fractures, and incapacity. Thus, anabolic therapy research is needed to increase physical function. Androgens show the most promise.

Selective androgen receptor modulators (SARMs) like Andarine and Ligandrol offer a more targeted approach to androgen therapy compared to traditional anabolic drugs [R]. SARMs selectively bind to androgen receptors (AR), influencing gene expression. This produces tissue-specific effects [R][R]. Unlike traditional drugs, SARMs exhibit reduced systemic side effects. This makes them promising candidates for research and potential therapeutic applications. Since AR is widely distributed, SARMs can target different tissues. These include increasing skeletal muscle growth, decreasing prostate size, and preventing breast cancer growth without systemic side effects [R][R].

Growth Hormone Secretagogues (GHSs) are sometimes used in research to study their combination with SARMs for potential anabolic effects [R]. MK-677 is a commonly studied GHS. It has shown promise as an anabolic agent. MK-677 may be researched in combination with SARMs like Andarine and Ligandrol.

To achieve lean mass and strength gains, researchers may consider combining Andarine and Ligandrol with MK-677. These compounds are often combined in research. This is done to potentially maximize their synergistic effects on muscle growth, strength, and fat loss.

S4 Andarine

Andarine is a selective androgen receptor modulator characterized by a comparatively short half-life [R]. It was originally formulated as a possible intervention for muscle atrophy, osteoporosis, and benign prostatic hyperplasia. SARMs provide a more precise methodology for androgen therapy. This has the potential to mitigate side effects in comparison to conventional pharmaceuticals. Andarine has exhibited anabolic effects in various studies. Nonetheless, the long-term safety and efficacy continue to be subjects of ongoing research. Andarine and other selective androgen receptor modulators have been shown to reduce muscle mass loss in clinical trials [R]. It has the potential to enhance bone retention, sexual function, and libido in women while minimizing the adverse effects commonly linked to conventional androgenic steroids [R].

How does S4 Andarine work?

S4 is a SARM that binds differentially to the AR. It alters receptor function upon binding [R]. Andarine mimics androgenic-anabolic steroids by attaching to androgen receptors. While reducing androgenic side effects, this induces muscle and bone protein anabolism [R].

SARMs have higher tissue selectivity for muscle and bone. This is compared to traditional steroid therapies, which significantly reduce side effects associated with androgenic tissues [R]. Unlike other drugs, SARMs are not converted by 5α-reductase and aromatase. This avoids the amplified androgenic and estrogenic effects in tissues like the prostate and seminal vesicles.

These kinds of effects are frequently seen when using testosterone and other anabolic medications [R]. Andarine is an agonist that promotes anabolism. When compared to anabolic-androgenic drugs, it has fewer side effects. This is mostly because it specifically targets 5α-reductase, the prostate’s enzyme that produces dihydrotestosterone (DHT) [R].

Research suggests that andarine may improve physical performance. These consist of skeletal health, strength, fat loss, and muscle growth. Andarine should still be used cautiously, even if it is thought to have a reduced risk profile than other drugs. Appropriate post-cycle therapy is recommended to minimize potential side effects.

S4 Andarine potential benefits:

• It may exert an anti-tumorigenic effect by promoting apoptosis. This inhibits the survival of cancer cells.
• It may reduce fracture risk and body fat by strengthening bones and reducing fall risk by strengthening muscles.
• It may improve muscle mass in chronic diseases.
• It is currently undergoing clinical development for the potential treatment of prostate cancer. [R]

What is Ligandrol?

Ligandrol, also known as LGD-4033, is a selective androgen receptor modulator. Its tissue selectivity and anabolic qualities make it a promising cachexia treatment. Research indicates that LGD-4033 is well-tolerated over chronic administration at various doses. Studies show that the compound consistently demonstrates androgenic activity. This is evident through an increase in lean body mass (LBM). It also shows notable suppression of testosterone and sex hormone-binding globulin. Additionally, it has been observed to lower HDL cholesterol levels [R].

A phase II clinical trial on Ligandrol, registered as VK5211, has been completed in the EU. This trial was randomized and double-blind. It included parallel-group, placebo-controlled, multicenter studies. Ligandrol studies were tested for efficacy, safety, & tolerability in acute hip fracture [R].

How it works

LGD-4033 is a highly selective non-steroidal SARM with a high binding affinity to AR. It acts as a weak agonist in the prostate [R]. Consequently, it is used for bulking due to its effect on improving muscle strength and hypertrophy. However, it does not cause androgenic effects like testosterone and other anabolic drugs. This helps prevent prostate hyperplasia [R].

The highly selective binding of Ligandrol is due to its high affinity (Ki of ~ 1 nM) to androgen receptors. LGD-4033 has produced muscle and bone anabolism and anti-resorption in animal models. It also shows robust selectivity for muscle versus prostate [R].

Ligandrol potential benefits:

• It may demonstrate a mild anabolic effect on the structural properties of osteoporotic bones.
• It may help improve hip fracture recovery in aged test subjects [R][R]
• It may aid in improving lean muscle mass, muscle strength, and sex hormones.
• It may potentially decrease prostate activity, which contributes to its enlargement. [R]
• It may act as a potential treatment for sarcopenia. [R]

What is MK-677?

MK-677, also referred to as ibutamoren mesylate or L-163,191, is a nonpeptide GHS. It is produced by the stomach and small intestine and can withstand the acidic conditions of the oral route [R]. MK-677 acts as a competitive agonist of Ghrelin. This allows it to stimulate various pathways influenced by the ghrelin receptor (GR) [R].

Ghrelin is the hunger hormone produced in the stomach. It acts as the natural ligand for the growth hormone (GH)-secretagogue receptor (GHS-R) [R]. This receptor could be an effective target for enhancing cognitive function and weight loss [R].

How does MK-677 work?

As an oral ghrelin mimetic, MK-677 can help manage short-term pre-meal hunger. Studies show it can increase GH secretion in test subjects without side effects. Additionally, MK-677 may help maintain fat-free mass and reduce abdominal visceral fat [R].

Once in the bloodstream, GH signals the liver to release the insulin-like growth factor 1 (IGF-1). IGF-1 triggers anabolic signaling in peripheral tissues, especially skeletal muscle. Additionally, activation of the GH receptor can initiate various downstream intracellular signaling pathways. These pathways may help improve the body’s overall structure [R].

MK-677 exhibits a half-life that is 12 times longer than Ghrelin. This longer half-life may boost GH and IGF-1 levels and efficacy [R]. These factors can significantly impact osteoblast formation [R].

MK-677 potential benefits:

• It may significantly address three key factors contributing to sarcopenia: reduced growth hormone secretion, loss of fat-free mass, and inadequate food intake [R].
• It may initiate a temporary weight loss effect and muscle building [R][R].
• This intervention may reverse protein degradation caused by calorie restriction, suggesting it may be useful in muscle loss cases [R].
• It potentially stimulates slow-wave sleep, resulting in an increased GH release. These may help improve the quality of sleep [R][R][R]
• It may accelerate bone turnover in older test subjects, increasing biochemical indicators of bone resorption and formation. [R]
• It may restore normal GH production to prevent signs of aging [R][R]
